the observed differences between the two methods. Our study is even more intriguing if we consider that, very recently, a new molecular form of PTH, with structural integrity of the PTH(1– 4) region and a modified PTH(15–20) region, has been identified by HPLC in primary and secondary hyperparathyroidism (21). In fact, this newly discovered form of PTH appears to be immu-noreactive and detectable by the whole-PTH assay but not by the intact-PTH assay. In conclusion, the unexpected constant differences in PTH values among immunoassays, observed in both uremic and PHPT patients and in healthy individuals, could be attributable to the different calibration procedures in addition to the presence of PTH fragments. It would be useful for manufacturers to reduce the systemic variability among methods by use of a more standardized method of calibration and use of antibodies that recognize the only biologically active PTH molecule. Accumulation of a non-(1– 84) molecular form of a parathyroid hormone (PTH) detected by intact PTH assay in renal failure: importance in the interpretation of PTH values. et al. Origin of parathyroid hormone (PTH) fragments detected by intact-PTH assays. non-(1– 84) circulating parathyroid hormone (PTH) fragment interferes significantly with intact PTH commercial assay measurements in uremic samples. New assays for intact parathyroid hormone and their clinical relevance for the diagnosis of hyperparathyroid-ism. Plasma levels of parathyroid hormone (1– 84) whole molecule and parathy-roid hormone (7– 84)-like fragments in pseudohypoparathyroidism type I. Development of a novel immunoradiometric assay exclusively for biologically active whole parathyroid hormone 1– 84: implications for improvement of accurate assessment of parathyroid function. Comparison of intact and " whole molecule " parathyroid hormone assays in patients with histologically confirmed post-renal transplant osteodystrophy.terminal form of parathyroid hormone (PTH) with immunologic similarities to hPTH(1– 84) is overproduced in primary and secondary hyperparathyroidism. Simultaneous multianalyte immunoassays offer advantages , including reduction of technical operations, execution time, and overall costs. The suspension array technology (SAT), which uses microfluorospheres with flow cytometry, is a multianalyte immunoassay that requires small specimen volumes, such as those obtained by less-invasive blood sampling approaches such as heel or finger sticks, as well as those obtained in pediatric specimens. The dried-blood-spot (DBS) technology has become an important screening tool for clinical and epidemiologic testing (1–3). It is particularly convenient in rural, resource limited settings where trained personnel and adequate facilities for blood collection, processing, transport, and storage may not be available. This approach, …

Determination of HIV-1 viral load on dried blood spot specimens
Dried blood spot specimens on filter paper were evaluated to determine the viral load of the human immunodeficiency virus type 1 (HIV-1), through the Cobas®Ampliprep/Cobas®Taqman® HIV-1 test. RNA levels were quantified in pairs of plasma and dry blood spot specimens on filter paper from persons infected with HIV-1. The Pearson linear correlation coefficient was calculated as well as the agreeme...

with the individual protein testing capacity of the WB. The assay can be tailored to the requirements of screening or confirmatory testing as well as to the needs of specific vaccine studies. Vaccine trials could benefit greatly from a PBA that differentiates protected from infected individuals. Because large-scale vaccine studies are likely to be conducted in settings where conditions may not ...
متن کاملApplication of dried blood spot specimens for serologic subtyping of human immunodeficiency virus type 1 in Thailand.
Dried blood spot (DBS) specimens were assessed as an alternative to plasma for human immunodeficiency virus type 1 (HIV-1) serotyping by V3 loop peptide enzyme immunoassay. Nested PCR capable of distinguishing HIV-1 subtypes B and E was used as the reference standard. Ninety-two percent of DBS samples were typeable as either HIV-1 subtype B or E. Serotype results with DBS and plasma were identi...
متن کاملAdaptation of the ultrasensitive HIV-1 p24 antigen assay to dried blood spot testing.
Implementation of molecular tests for the assessment of pediatric HIV-1 infection in resource-limited countries is difficult because of technical complexity and costs. Alternatives like the ultrasensitive HIV-1 p24 antigen enzyme-linked immunosorbent assay have therefore been proposed. We have now adapted this test to dried blood spot (DBS) plasma p24 antigen (p24). High background activity was...

Background/Objective: Development of a new enzyme-linked immunosorbent assay (ELISA) for screening human blood serum and plasma for antibodies to human immunodeficiency virus type 1 (HIV1) and type 2 (HIV2) as HIV1/2REC ELISA diagnostic kit based on E. coli derived soluble recombinant proteins. Methods: Polypeptides corresponding to HIV1 gp41 and HIV2 gp36 immunodominant regions and HIV1 gag we...
